Start with building a part list. As it will say on most websites, do your own research when it comes to picking parts, but a good place to get a good idea where to start would be heading over to:
logicalincrements.com
examine the parts in your budget ammount and keep those in mind. Then go to:
pcpartpicker.com
and check out their huge database of parts. When you find out what works for YOUR needs, and not the needs of others make a build on pcpartpicker to check for discrepancies in your selected parts. These discrepancies can range from the wrong form-factor between your motherboard and case or even if your Video Card won't fit correctly. Once you have no discrepancy head over to either:
Amazon.com
newegg.com
tigerdirect.com
or even a local brick and mortar store.(They are unlikely to stock EVERY part possible so better to order online.)
Once you order your parts you have a variety of options. You can use the motherboard manual that will explain how to build your computer step-by-step or you can check out one of the hundreds of online sources that will walk you through building a computer step-by-step. Some examples of websites would be:
