As an experienced DM, I can only say that you really need to be comfortable with going on the fly.

You have to be sure that what ever it is that you have to come up with, isn't clunky, disorganized, or inane. What seems like a good idea in your head can, mechanically, be a pain in the ass.

write up a LOOSE plot. try to keep your players on a plot with pre-made NPC's but if they stray too far away from your plot, rework it to fit their wants. Nobody likes a railroading GM.

With that in mind, some players need a little railroading, usually through negative reinforcement. Give them a spank if they spend to long harassing barmaids and not slaying goblins.

I'm running a DW campaign "because it's easy" and about to start a Dark heresy campaign with an entirely different group.

Mixture of stoked and stressed. Especially since I'm not extremely adept at Roll20 yet.

I suggest not having something as large in scope as an inquisitorial investigation just yet. Perhaps some Arbites who start finding signs of heresy and stumble across a larger cult?
It has less scale but it follows much the same themes and allows for a more railroaded plot.

I've already got the grand plot done, actually. It's pretty extreme and badass near the end, but I plan on making them work and most likely die for it.

But yeah, starting out small would be the best plan. My only real issue is how to make the Inquisitor interact with the Acolytes. How do they get their orders/get around, etc. Public transport I'd imagine?

I've gotta say, Terra is way too damn pure for anything bad to happen at all. As in, with it being the place where the Big E himself is situated, nobody in their right mind will allow one micrometer to be unchecked for HERESY. I would suggest a particularly massive hive world, plenty of transit tunnels and trains to get around, populations numbering in the tens of billions (large ones averaging around 50 billion), and some real shady shit happening in the lower levels. Lots of room for heresy'n'shit.

Oh certainly not Terra itself. I just figured that would be a good "Tavern" so to speak. A place for these random acolytes to get assembled and assigned a task by their Inquisitor.

A Hive world would work well. I'm thinking of starting out with simply seditionists or traitors that aren't necessarily chaos corrupted.

We have a Krieg, so that is sure to work out well.

B) low profile, in a hive when word gets out that some =][= acolytes are about, the heretics hush up a bit.

Oh, i look forward to hearing how a Krieger does. But i would say have the big =][= contact them remotely, maybe give them a long-range vox caster (backpack sized?) to contact the inquisitor from another place on the planet or maybe a small ship in orbit?

The idea is that this Inquisitor isn't really on hand. He's a busy man, etc etc. Wouldn't show up for some seditionists when he could throw a team of Acolytes at it and clean up afterwards if they fail.

If you like, I could try and keep you posted. He's basically a Vatt-born Krieg with mild mutations. Red eyes and left handed, and over-excelled in training because of the harsher treatment. Plucked up by the inquisition when an agent saw his record of service. He would have been squandered in the Death Corps, due to his mutations.

We also have a Pskyer, an Adept, and a Tech-Priest that lack a solid background yet,

and a Arbitraror who was sold to tech-priests as a child, and eventually got landed on a feral world where he was raised.

If you'd like more details of the /actual/ plot, let me know. I'd be happy to get some outside ideas on it and such. Be warned, it's got some pretty mega-powerful shit in it near the end.
