well in the simplest form, it is about a madman with a big blue box.

now to elaborate on that it is about the Doctor, who is Gallifreyan, or a Time Lord, the last of an extremely advanced species that is extremely similar to human in physical features, save for 2 hearts. He travels through time and space via the TARDIS (Time and Relative Dimensions in Space) whenever he senses that there is trouble that he needs to attend to, whether it be fighting off the Daleks, an extremist species hell-bent on exterminating any lesser species, and to the, all other species are lesser then them, to saving the universe and restarting the Big-Bang to vacationing on a planet with 2 time streams separated by only an elevator. The Doctor also has the ability to regenerate when he sustains fatal injuries, reincarnating near instantly in a new body with a new sense of style, taste and behavior, though t=still at the core remaining The Doctor. The show itself is very, very enjoyable though needs to be watched in sequence. The Doctor himself, in his current form, the 11th doctor loves Bowties, Fezes, Stetsons, Jammie-dodgers and hates beans, bread-and-butter, bacon and apples.
He picks up numerous companions throughout his various incarnations and is witty and quirky in various ways with each incarnation, such as the 9th's doctors near obsessive love of bananas or the 10th's quirk of wearing lense-less glasses to make himself feel smarter and way of explaining time as a 'Big ball of wibbly-wobbly, timey-wimey...stuff." the 11th's tendency to go on and on rambling quickly coming up with the problems and quickly ditching the solution in a matter of seconds and pacing, as well as his cartoonishly exuberent flourishes and somewhat naive nature when he is not trying to be serious as well as his completely random sense of driving the tardis, pacing and spinning around the control panel, flipping switches and turning knobs at whimsy and yet somehow managing to get where he wants to go.

My nerd senses are tingling to correct you on a few things.
1. He has only been the last Time Lord after he locked his entire race in a time lock, sometime in his 8th or 9th incarnations.
2. It's Time and Relative Dimension in Space (singular 'Dimension')
3. The vast majority of the show's run (yes, even the new ones) are in self-contained stories and do not need to be watched in sequence. It's only really since Moffatt that that has really mattered.
