Midi-chlorians don't actually cause the force, they are sentient microorganisms that symbiotically feed off the force in their host. The stronger you are in the force, the more midi-chlorians live in you.

I understand that an extra chromosome in the last pair of 24 chromosomes (altogether 48 chromosomes) would create down syndrome (when someone has 49 chromosomes.)
Is that the joke?

In Star Wars Episode 1, they introduce this concept called "Midi-chlorians" which are something in the blood that gauge a person's force capabilities (the fact was later edited and corrected, thankfully, in the extended universe). When Qui-Gon-Jin took Anakin's blood, he said he had a number of midi-chlorians that was off the charts.
In this comic, he says he got it confused, and Anakin has more chromosomes than anyone before, rather than midi-chlorians.

also down syndrome is caused by an extra 21st chromosome, not the final chromosome, that would be an extra sex chromosome, and depending on the gender and which chromosome is duplicated, could cause a variety of other disorders
