In Starcraft, the various races compete to gather resources known as minerals and vespene gas, which they then use to create armies to destroy each others bases. While minerals can be harvested by the workers without anything to assist them, vespene gas can only be harvested from vespene geysers, which in turn require a specific structure (in the case of the Terrans, the refinery) before harvesting can be commenced. While vespene gas is essential for late game units and structures, it has no function in the opening minutes of a game. Therefore, to build one wastes minerals which could be used to produce more workers or structures you require, and takes workers away from the more essential resource in the early game to harvest a resource not necessary at all in the first five minutes. Contrast this with what our character, Derp, clearly did, along with the title of the comic 'Starcraft Pro', and you are able to see where the humor in the joke comes from (in this case, irony, and the image of what is implied to be a more experienced Starcraft player laughing at Derp.)

tl;dr. Derp is a noob, laugh at him.
