Sorry, I didn't make myself clear. They're only brothers in the first anime, but that is a spin-off from the manga. It says in the manga and FMA:Brotherhood that the homunculi are genderless creatures.

That and in the spinoff envy shows her true form as a blonde dude. In brotherhood her true form is a tiny dinosaur like creature
