He's actually not wrong. In photography, film and graphic design there's a "rule" of composition known as the Rule of Thirds.
Essentially you take the frame and divide it in thirds vertically and horizontally. You're supposed to try to position the subject(s) of the image on the lines and/or intersections. It doesn't have to be precise and it's also not a rule for every photo, just a "guide" or "suggestion" so to speak.
If you take the individual frames of the comic and apply the rule, you'll see it's pretty much dead on.
I'm sure it is a joke, but any photograhper or graphic designer worth his salt knows of this rule. If the creator of these comics studied GD it's likely part of the reason it's so recurring in his comics.
The four panel format has been common since literally the beginning of comics. It's irrelevant whether or not there was a reason behind it originally, I can guarantee that the guy doesn't know a thing about it.

I don't know a thing about the artist as to rebut your guarantee, but unless you know him personally or it's well known that he's never looked into graphic design, you are making a very bold statement.
First off, the meme can attest that the guy's an idiot. Second, the only possible thing you could have been talking about was the four panel format, since the only characters that are small enough to be placed anywhere but the intersections aren't anywhere near them. Third, the lines in >>#26 are clearly just there to emulate the characters in the first place, not to indicate any kind of photography technique; if you were unaware, the lines were added well after the comic was created by someone meaning to mock the original artist.

I noticed that the lines are on the characters, I'm not blind. In fact that's what assholy was pointing out: within each panel, the characters are approximately on the thirds. I never said the lines were part of the comic, as I said: "I'm talking about the position of the characters within each panel."
Then again I'm talking to someone who thinks memes are proof of a person's intelligence.
